What Is DNA

DNA, or deoxyribonucleic acid, is the instruction manual inside almost every cell of every living thing. It tells cells how to grow, repair themselves, behave, and function. Every feature of your body is stored inside your DNA.

Most of the time DNA is far too small to see. It is tightly coiled inside the nucleus of each cell. But when we break open the cheek cells in your mouth and separate the DNA from everything else, the strands clump together and rise into view.

That is the magic of this experiment.

Why Human Cheeks

Your mouth is one of the easiest places to collect human cells because:

• Small bowl

• Optional: pipettes, test tubes, or droppers
• Optional: goggles, apron, notebook

Everything else you need is already in your cheeks.

Step-by-Step Instructions

1. Make Your Salty Mouth Rinse

In a cup, mix:

• Half cup warm water
• Two teaspoon salt

Warm water helps dissolve the salt completely.

Salt helps loosen cheek cells from the inside of your mouth.

Stir until the water is clear.

2. Collect Your Cheek Cells

Take a sip of your salty water and swish it in your mouth for 2 minutes.
Swish with gentle force so you dislodge many cells.

Do not swallow.
Spit the water back into your cup.

Your cheek cells are now floating inside this salty liquid.

3. Add the Extraction Solution

In a separate bowl, mix:

• One teaspoon dish soap
• Cheek and salt mix solution

Stir very gently.

Dish soap dissolves the membranes of your cheek cells so the DNA can escape.

Pour this mixture into your cup of cheek-cell water.

Let it sit still for one minute.

4. Add the Cold Alcohol

Tilt the cup slightly and gently pour the cold rubbing alcohol down the side so it forms a clean top layer.

Do not stir.

5. Watch Your DNA Appear

Within a three to five minutes, cloudy white strands will begin rising into the alcohol layer.

This wispy, thread-like material is human DNA.
