Extracting DNA from Human Cheeks: A Homeschool Science Adventure
A Hands-On Way To Discover The Instructions Inside You
When you think of DNA, you might picture a high-tech laboratory with scientists moving between microscopes, glass tubes, and glowing monitors. But what if I told you that you carry all the ingredients to extract DNA right inside your own mouth, and you can pull it out in your kitchen with a few simple tools and a sprinkle of Armitage House magic?
Human DNA extraction is one of the most exciting experiments you can do at home. It is safe, simple, and absolutely unforgettable for children and adults. It turns the invisible into something you can actually see. And at Armitage House, we believe that when a child can hold science in their hands, something powerful awakens inside them.
Let the adventure begin.
What Is DNA
DNA, or deoxyribonucleic acid, is the instruction manual inside almost every cell of every living thing. It tells cells how to grow, repair themselves, behave, and function. Every feature of your body is stored inside your DNA.
Most of the time DNA is far too small to see. It is tightly coiled inside the nucleus of each cell. But when we break open the cheek cells in your mouth and separate the DNA from everything else, the strands clump together and rise into view.
That is the magic of this experiment.

Why Human Cheeks
Your mouth is one of the easiest places to collect human cells because:
• You naturally shed thousands of cheek cells every minute
• They come off easily with a little water
• They contain plenty of DNA
• They are safe, free, and accessible for homeschooling families
• Children love the idea of extracting DNA from themselves
This experiment helps kids connect science to their own bodies. It makes biology personal, relatable, and exciting.
Materials You Will Need
• Two tablespoons salt
• Half cup warm water
• One teaspoon soap dish
• Clear glass or cup
• Rubbing alcohol chilled in the freezer for at least one hour
• Small bowl
• Spoon
• Measuring spoons
• Optional: food coloring for visibility
• Optional: pipettes, test tubes, or droppers
• Optional: goggles, apron, notebook
Everything else you need is already in your cheeks.
Step-by-Step Instructions
1. Make Your Salty Mouth Rinse
In a cup, mix:
• Half cup warm water
• Two teaspoon salt
Warm water helps dissolve the salt completely.
Salt helps loosen cheek cells from the inside of your mouth.
Stir until the water is clear.
2. Collect Your Cheek Cells
Take a sip of your salty water and swish it in your mouth for 2 minutes.
Swish with gentle force so you dislodge many cells.
Do not swallow.
Spit the water back into your cup.
Your cheek cells are now floating inside this salty liquid.
3. Add the Extraction Solution
In a separate bowl, mix:
• One teaspoon dish soap
• Cheek and salt mix solution
Stir very gently.
Dish soap dissolves the membranes of your cheek cells so the DNA can escape.
Too much stirring creates bubbles, so go slowly.
Pour this mixture into your cup of cheek-cell water.
Let it sit still for one minute.
4. Add the Cold Alcohol
Tilt the cup slightly and gently pour the cold rubbing alcohol down the side so it forms a clean top layer.
Do not stir.
This moment feels like the beginning of magic.
5. Watch Your DNA Appear
Within a three to five minutes, cloudy white strands will begin rising into the alcohol layer.
This wispy, thread-like material is human DNA.
Your DNA.
Right in front of your eyes.
Children never forget this moment.
The Science Behind The Magic
• Salt loosens cheek cells inside your mouth
• Soap breaks open the cell membranes
• The mixture releases DNA into the water
• Alcohol forces the DNA to clump together
• Human DNA becomes visible as white cloudy strands
Scientists use the same basic principles in real genetics labs. We simply adapt the steps to be safe and kitchen-friendly.
Learning Extensions and Wonder Activities
1. Compare Yourself To Fruit
Extract DNA from bananas or strawberries, then compare it to your cheek DNA.
Which one produced more?
Create a chart.
2. Creative Storytelling
Write from the perspective of a cheek cell:
“I lived on the inside of your mouth until today…”
Encourage imagination, emotion, and humor.
3. Build a DNA Model
Use pipe cleaners, beads, or candy to form a double helix.
4. Microscopic Art
Draw what you saw and label:
• cheek cells
• DNA strands
• alcohol layer
5. Talk About Inheritance
Why do children sometimes look like their parents?
Why can two siblings look different?
Open the door to genetics naturally.
Why This Experiment Matters At Armitage House
At Armitage House, we believe that science is not memorization. Science is wonder. Science is play. Science is creativity.
Children learn best when:
• they touch
• they see
• they feel
• they experiment
• they ask questions
• they make connections between real life and big ideas
Extracting DNA from human cheeks makes biology meaningful. It builds confidence. It encourages curiosity. It helps children understand that learning happens inside their own bodies.
This experiment is an unforgettable introduction to genetics and a boost for imagination, creativity, and wonder-driven learning.
